ABSTRACT

Objective:This study aims to observe the effect of baicalein on the clonal formation of triple negative breast cancer MDA-MB-231 and MDA-MB-468 cells, and to explore the mediation role of Yes- related protein (YAP) in it. Method:MDA-MB-231 and MDA-MB-468 cells were treated with baicalein. Thiazole blue (MTT) colorimetric method was used to detect cell proliferation ability. Plate cloning experiments was used to detect the colony forming ability. Immunofluorescence method was used to detect the nuclear distribution of YAP, and Western blot test was used to detect the protein expression levels of YAP large tumor suppressor factor 1 (LATS1), YAP, phosphorylated Yes- related protein(p-YAP) and phosphorylated YAP large tumor suppressor factor 1 (p-LATS1). Result:Compared with the blank group, baicalein (40, 80, 160 μmol·L<sup>-1</sup>) significantly inhibited the proliferation ability of MDA-MB-468 and MDA-MB-231 cells (<italic>P</italic><0.05, <italic>P</italic><0.01), and the inhibitory effect was dose-dependent. The half inhibit concentration(IC<sub>50</sub>) of baicalein against MDA-MB-468 and MDA-MB-231 cells were (80.3±7.2),(70.4±6.5) μmol·L<sup>-1</sup>, respectively. Compared with blank group, baicalein (5, 10, 20 μmol·L<sup>-1</sup>) had no significant effect on the proliferation of MDA-MB-468 and MDA-MB-231 cells, and the difference was not statistically significant. Compared with the blank group, baicalein (5, 10, 20 μmol·L<sup>-1</sup>) significantly dose-dependently reduced the cell colony formation rates of MDA-MB-468 and MDA-MB-231 cells (<italic>P</italic><0.05, <italic>P</italic><0.01), and baicalein (10, 20 μmol·L<sup>-1</sup>) significantly inhibited the nuclear expression of YAP in MDA-MB-468 and MDA-MB-231 cells in a dose-dependent manner(<italic>P</italic><0.01). Also, baicalin (5, 10, 20 μmol·L<sup>-1</sup>) significantly up-regulated p-YAP and p-LATS1 protein expressions in MDA-MB-468 cells in a dose-dependent manner (<italic>P</italic><0.05, <italic>P</italic><0.01). Baicalein (10, 20 μmol·L<sup>-1</sup>) significantly up-regulated p-YAP and p-LATS1 protein expressions in MDA-MB-231 cells in a dose-dependent manner (<italic>P</italic><0.01). Conclusion:Baicalein can inhibit colony formation of triple negative breast cancer MDA-MB-468 and MDA-MB-231 cells by mediating the reduction of YAP entry into the nucleus.

ABSTRACT

Objective: To analyze the influence of serum high mobility group box-1 protein (HMGB1) levels on the severity and prognosis of critical ill patients at the early of trauma-induced coagulopathy (TIC) in intensive care unit (ICU). Methods: 43 cases of critical ill patients with severe trauma were included during January 1, 2017 to December 31, 2018 in ICU of Foshan Hospital of Traditional Chinese Medicine. International normalized ratio (INR) >1.2 was applied as the diagnosis criterion of TIC. The patients was divided into TIC group (n=23) and control group (n=20). Their age, sex, injury mechanism, the interval between injury and admission to ICU (delay time), the interval between injury and coagulopathy correction (correction time), ISS scores, APACHE II scores at admission to ICU were recorded, and the activated partial thromboplastin time (APTT), prothrombin time (PT), INR, fibrinogen (Fib), platelet counts (PLT), C-reactive protein (CRP) and procalcitonin (PCT) levels were detected meanwhile. The serum HMGB1 levels were examined through ELISA. The blood transfusion volume (red blood cells, RBC and fresh frozen plasma, FFP), ventilation time, ICU stay and 28-day mortality rate were statistically analyzed. Results: TIC occurred in 53.5% of critical ill trauma patients in ICU. There were no significant differences in the age, sex, injury mechanism, delay time, APACHE II scores, ISS scores, APTT, PT, CRP and PCT levels between two groups (P>0.05). Compared with control group, the Fib and PLT levels were significantly reduced in the TIC group, and the ventilation time, blood transfusion volume of RBC and FFP, infection rate and organ dysfunction rate were remarkably increased (P<0.05). Besides, the 28-day mortality rate revealed a raised tendency in TIC group (P=0.091). Simultaneously, the serum HMGB1 levels at admission to ICU were significantly increased in the TIC group, and the serum HMGB1 level in the death subgroup was much higher than that in the survivors, as same as those in the TIC subgroup analysis (P=0.000). The correlation analysis disclosed that serum HMGB1 levels at admission was positively related to delay time, correction time, APACHE II score, ISS score, ventilation time, INR levels, APTT, CRP and PCT levels at admission (r=0.648, 0754, 0.526, 0.516, 0.521, 0.509, 0.432, 0.592, 0.375), and was negatively associated with Fib levels, PLT value, infection incidence rate, organ dysfunction rate and 28-day mortality rate (r=-0.424, -0.571, -0.505, -0.396, -0.765). There were significant differences in the delay time, correction time, ISS scores, transfusion volume, serum HMGB1 levels, PLT value, APTT and CRP levels between death subgroup and survivor subgroup of TIC patients (P<0.05), and serum HMGB1 levels and PLT value at admission were independent risk factors in the multivariable logistic regression analysis (P=0.004, 0.011). For the TIC sub-group trauma patients, the AUC of serum HMGB1 levels was 0.897(95%CI 0.748-1.000, P=0.002), the best cutoff value was 54.60 ng/ml with Youden index of 0.808. Conclusions: The PLT levels and serum HMGB1 levels at admission to ICU are independent risk factors of critical ill patients with severe trauma. The serum HMGB1 levels is closely related to severity and prognosis, and can predict the 28-day mortality rate of critical ill patients with TIC.

ABSTRACT

To explore the optimal therapy time for the treatment of severe coronavirus disease 2019(COVID-19)by traditional Chinese medicine(TCM)and its influence on the therapeutic effect and prognosis. The clinical data,laboratory findings,and outcomes of 64 patients with severe COVID-19 treated with TCM and western medicine in Chongqing from January 20,2020, to March 11,2020 were retrospectively analyzed.Patients were divided into early intervention group[TCM was initiated within 3 days (including day 3) after the first diagnosis of severe type/critical type COVID-19]and late intervention group[TCM was initiated after 7 days (including day 7) after the first diagnosis of severe type /critical type COVID-19].The changes in clinical parameters during the course of disease were compared between the two groups. On day 14,the oxygenation index was 292.5(252.0,351.0)mmHg in the early intervention group,which was significantly higher than that in the late intervention group [246.0(170.0,292.5)mmHg](=0.005).The length of hospital stay [(18.56±1.11)d (24.87±1.64)d,=0.001],duration of ICU stay [(14.12±0.91)d (20.00±1.53)d,=0.000] and time to negativity [(16.77±1.04)d (22.48±1.66)d,=0.001] in the early intervention group were significantly shorter than those in the late intervention group.The intubation rate(7.3%)in the early intervention group was significantly lower than that in the late intervention group(30.4%)(=0.028). Early TCM therapy within three days after a diagnosis of severe COVID-19 can shorten the length of hospital stay,duration of ICU stay,and time to negativity and decrease intubation rate.

ABSTRACT

Objective@#Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.@*Methods@#A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients' medical records. Multivariable logistic regression models were used to estimate the odds ratio ( @*Results@#Overall, 158 (13.6%) patients were diagnosed with severe illness and 32 (2.7%) had unfavorable outcomes. Hypertension (2.87, 1.30-6.32), type 2 diabetes (T2DM) (3.57, 2.32-5.49), cardiovascular disease (CVD) (3.78, 1.81-7.89), fatty liver disease (7.53, 1.96-28.96), hyperlipidemia (2.15, 1.26-3.67), other lung diseases (6.00, 3.01-11.96), and electrolyte imbalance (10.40, 3.00-26.10) were independently linked to increased odds of being severely ill. T2DM (6.07, 2.89-12.75), CVD (8.47, 6.03-11.89), and electrolyte imbalance (19.44, 11.47-32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission (5.46, 3.25-9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes (6.58, 1.46-29.64) within two weeks.@*Conclusion@#Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. Women with comorbidities were more likely to have severe disease, while men with comorbidities were more likely to have unfavorable treatment outcomes.

ABSTRACT

Objective:To observe the effect of Liuwei Dihuangtang on the mitochondrial apoptotic pathway in rats with chronic renal failure. Method:The 45 male SD rats were randomly divided into normal group (n=10), sham group (n=10) and operation group (n=25). The operation group received 5/6 nephrectomy, and the rats in operation group were randomly divided into the model group and the treatment group after successful modeling. The treatment group received Liuwei Dihuangtang by gavage administration, while the rest of the three groups received equal volume of distilled water by gavage administration, with a treatment course of 8 weeks. The quantitative levels of serum creatinine (SCr), blood urea nitrogen (BUN) and urine protein in each group were determined. Hematoxylin eosin (HE) staining and Masson staining were used to observe the histopathological changes of rat kidney. The structural changes of mitochondria in renal tubular epithelial cells were observed under electron microscope. The apoptotic rate was detected by terminal-deoxynucleoitidyl transferase mediated nick end labeling(TUNEL). The expression of B cell lymphoma -2 (Bcl-2), Bcl-2 associated X protein (Bax), cytochrome C (Cyt C) and cysteine aspartic acid protease -3 (Caspase-3) were analyzed by Western blot and Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR). Result:As compared with the normal group, the quantitative levels of SCr, BUN and urinary protein in the model group were significantly increased (P<0.01), and the above indicators were improved in the treatment group as compared with the model group (P<0.05). No significant pathological changes and damage of mitochondrial structure were observed in the kidney tissues of the control group and the sham group. In the model group, there were different degrees of renal tubular atrophy and dilatation, interstitial inflammatory cell infiltration and collagen deposition, and mitochondria swelling and breaking, but such lesions were significantly alleviated in the treatment group. As compared with the normal group, apoptotic rate was increased significantly in model group (P<0.01), the expression of Bax and Caspase-3 was significantly increased (P<0.01), and the expression of Bcl-2 was significantly decreased (P<0.01). As compared with the model group, apoptotic rate was significantly reduced in the treatment group (P<0.01), the expression of Bax and Caspase-3 was significantly decreased (P<0.01), and the expression of Bcl-2 was significantly increased (P<0.01). In the normal group and sham group, Cyt C was mainly located in mitochondria. The expression of Cyt C in the cytoplasm of the model group was increased significantly (P<0.01), while that of the treatment group was decreased (P<0.01). Conclusion:Liuwei Dihuangtang can alleviate renal fibrosis and delay the progression of renal function. The mechanism of reducing apoptosis may be associated with protecting mitochondrial structure and regulating the expression of proteins related to mitochondrial apoptotic pathway.

ObjectiveUsing Chromium-51 release assay, lactate dehydrogenase release assayand other methods to detect the cytotoxicity of CD19 CAR-T cells is cumbersome, with low repeatability and poor stability. This study aims to establish a label-free and real-time method for detectingspecific cytotoxicity of CD19 CAR-T cells.MethodsIn order to establish target cell models for cytotoxic assay of CD19 CAR-T cells by using Real Time Cellular Analysis (RTCA) system,the adherent human breast cancer cells were infected with lentiviral vectors encoding CD19. CD19 expression on the transduced cells was detected by flow cytometry. The cellsexpressing CD19 stably werethen sorted by fluorescence activated cell sorting (FACS).With such cells as target cells, CD19 CAR-T cells and BCMA CAR-T cells as effector cells, RTCAsystem was used to evaluate the cytotoxicity of CAR-T cells against target cells.ResultsMDA-MB-231 and SKBR3cells with stable expression CD19were obtained in this study.The results of flow cytometry showed that positive expression rate of CD19 in MDA-MB-231/CD19 cells and SKBR3/CD19 monoclonal cells were 99.03% and 98.91%,respectively.RTCA results showed that with MDA-MB-231 and MDA-MB-231/CD19 cells as target cells,CD19 CAR-T cells showed significant cytotoxicity to MDA-MB-231/CD19 cellsat the effector-target ratio of 5∶1, 1∶1 and 1∶5,but not to MDA-MB-231 cells. With SKBR3 and SKBR3/CD19 cells as target cells, CD19 CAR-T cells showed significant cytotoxicity to SKBR3/CD19 cellsat the effector-target ratio of 5∶1and 1∶1. When the effector-target ratio was 1∶5, there was no obvious cytotoxicity.The data of MDA-MB-231/CD19 or SKBR3/CD19 as target cells and CD19 CAR-T as effector cells were analyzed separately, showing that when the number of target cells was the same, the cytotoxicity detected by RTCA increased as the number of CD19 CAR-T cells increased.The cytotoxic assays of CD19 CAR-T cells showed specificity and dose-response relationship of CD19 CAR-T cytotoxicity against the target cells.ConclusionThis study established a method for evaluating cytotoxicity of CD19 CAR-T cells that is real-time, label-free, simple and convenient.

ABSTRACT

Objective To investigate the effects of Panax Notoginseng saponins (PNS) on protein expression of Klotho in rats with renal ischemia reperfusion injury; To discuss its protective mechanism for model rats. Methods Experimental rats were randomly divided into sham-operation group, model group, positive medicine group, PNS high-, medium- and low-dosage groups. Each administration group was given relevant medicine for gavage, once a day. Renal ischemia reperfusion injury model was established. Rats were sacrificed by taking blood from abdominal aorta after 4 hours of modeling. Serum levels of blood urea nitrogen (BUN), creatinine (SCr), malondialdehyde (MDA) content in kidney tissue, superoxide dismutase (SOD) activity and glutathione peroxidase (GSH-Px) activity were measured. HE staining was used to observe the morphological changes of renal tissue. The protein expressions of Klotho and NF-κB p65 were measured by immunohistochemical method. Results Compared with the sham-operation group, the levels of BUN and SCr in the model group increased significantly (P<0.05); protein expression of Klotho in renal tissue decreased and the protein expression of NF-κB p65 increased (P<0.05). Compared with the model group, the expression of Klotho increased but protein expression of NF-κB p65 decreased in each administration group (P<0.05); Compared with the positive medicine group, the expression of Klotho in PNS high-dosage group increased but protein expression of NF-κB p65 decreased (P<0.05). The protein expression of NF-κB p65 was negatively related to protein expression of Klotho (r=-0.895, P<0.05). Conclusion PNS can inhibit oxidative stress and anti-inflammatory effects through upregulating protein expression of Klotho, and reduce the protein expression of NF-κB p65, and thus exerts renal protective effects.

ABSTRACT

BACKGROUND:Stellate ganglion block is feasible for the patients in sub-health status,but it is invasive and the patient compliance is poor.OBJECTIVE:To investigate the clinical effectiveness of ultrasound-induced transdermal drug delivery for reducing the hyperactivity of the cervical sympathetic ganglia in the sub-health status.METHODS:Sixty-nine participants in sub-health state from different age levels and professions were recruited and were randomly divided into treatment (n=31) and control (n=38) groups.The treatment group underwent ultrasound-induced transdermal drug delivery to the cervical sympathetic ganglia,while the control group received psychological counseling on sub-health education and behavior intervention.All patients were assessed with Sub-Health Measurement Scale Version1.0,and the Medical Outcomes Study 36-Item Short-Form Health Survey prior to and post treatment,along with clinical curative effect assessment.RESULTS AND CONCLUSION:After treatment,29 participants in the treatment group were improved or recovered from sub-health,with an effective rate of 93.5%.Compared with the control group,the scores were significantly improved in the treatment group.To conclude,ultrasound-induced transdermal drug delivery to the stellate ganglion has a significant effect on sub-health state.

ABSTRACT

The present study was designed to determine the effects of Guanfu base A (GFA) on the late sodium current (INa.L), transient sodium current (INa.T), HERG current (IHERG), and Kv1.5 current (IKv1.5). The values of INa.L, INa.T, IHERG and IKv1.5 were recorded using the whole-cell patch clamp technique. Compared with other channels, GFA showed selective blocking activity in late sodium channel. It inhibited INa.L in a concentration-dependent manner with an IC50 of (1.57 ± 0.14) μmol · L(-1), which was significantly lower than its IC50 values of (21.17 ± 4.51) μmol · L(-1) for the INa.T. The inhibitory effect of GFA on INa,L was not affected by 200 μmol · L(-1) H2O2. It inhibited IHERG with an IC50 of (273 ± 34) μmol · L(-1) and has slight blocking effect on IKv1.5, decreasing IKv1.5 by only 20.6% at 200 μmol · L(-1). In summary, GFA inhibited INa.L selectively and remained similar inhibition in presence of reactive oxygen species. These findings may suggest a novel molecular mechanism for the potential clinical application of GFA in the treatment of cardiovascular disorders.

ABSTRACT

Objective To discuss the mechanism of Liuwei Dihuang Decoction against renal interstitial fibrosis. Methods The rat models of renal interstitial fibrosis were induced by 5/6 nephrectomy, and were randomly divided into sham-operation group, model group, Liuwei Dihuang Decoction group and enalapril group. Liuwei Dihuang Decoction group was given 6.25 g/(kg?d) of Liuwei Dihuang Decoction for gavage, enalapril group was given 10 mg/(kg?d) enalapril for gavage, sham-operation group and model group were given 10 mL/(kg?d) of distilled water for gavage, 1 time per day for 12 weeks. Expressions of HIF-1α, CollI, and CollIII in kidney tissue were detected by immunohistochemistry. Results Compared with the sham-operation group, the expressions of HIF-1α, CollI, and CollIII of renal tissue in the model group significantly increased (P<0.05). Compared with the model group, Liuwei Dihuang Decoction group and enalapril group can inhibit the expressions of HIF-1α, CollI, and CollIII (P<0.05), and Liuwei Dihuang Decoction group was better than enalapril group (P<0.05). Conclusion The mechanism of Liuwei Dihuang Decoction against renal interstitial fibrosis may improve the renal tissue of chronic hypoxia to play the role of preventing renal interstitial fibrosis by decreasing the expressions of HIF-1α, CollI, and CollIII.

ABSTRACT

Objective To observe the effect of Chinese yam polysaccharide on the expression of hypoxia inducible factor-1α (HIF-1α) and vascular endothelial growth factor (VEGF) in rats with renal ischemia reperfusion injury. Methods SD rats were randomly divided into sham operation group, model group and Chinese yam polysaccharide group. Renal ischemia reperfusion injury model in rats was prepared. Seven days before operation, Chinese yam polysaccharide group was gastrically administrated with yam polysaccharide (200 mg/kg) daily, sham operation group and model group with same volume of saline respectively. BUN and SCr were detected postoperative 6 h, the protein expression of HIF-1αin renal tissue was detected by Western Blot, and HIF-1αand VEGF mRNA expression in renal tissue were detected by RT-PCR. Results Compared with sham operation group, serum levels of BUN and SCr of model group and Chinese yam polysaccharides group increased (P<0.01), and the mRNA and protein expression of HIF-1α, mRNA expression of VEGF in renal tissue increased (P<0.01). Compared with model group, serum levels of BUN and SCr of Chinese yam polysaccharide group decreased (P<0.01), and HIF-1α mRNA and protein expression, VEGF mRNA expression in renal tissue increased (P<0.01). Conclusion Chinese yam polysaccharide has obvious protective effect on renal ischemia reperfusion injury, which may be related with up-regulation of HIF-1αexpression and induction of VEGF expression.

AIM@#In a search for new cardiovascular drug candidates, a series of novel oxime ethers derived from a natural isochroman-4-one were synthesized.@*METHOD@#Compounds 3 and 6, derived from the natural antihypertensive compound 7, 8-dihydroxy-3-methyl-isochroman-4-one (XJP), were designed and synthesized. Subsequently, a series of novel isochroman-4-one oxime ether hybrids were prepared by hybridizing various N-substituted isopropanolamine functionalities to isochroman-4-one oxime. Furthermore, β1-adrenergic blocking activities of the synthesized compounds were assayed using the isolated rat left atria.@*RESULTS@#Twenty target compounds were obtained, and the preliminary structure-activity relationships were deduced. The most promising compound Ic exhibited β1-adrenoceptor blocking activity (inhibition: 52.2%) at 10(-7) mol·L(-1), which was superior to that of propranolol (inhibition: 49.7%).@*CONCLUSION@#The results suggested that natural product XJP/isopropanolamine moiety hybrids may provide a promising approach for the discovery of novel cardiovascular drug candidates.

This paper explored influences of situational teaching competition on improvement of pathology teaching effect.Students were grouped to create situational teaching works under teacher's guidance and finally competition was carried out.Teachers were invited to do the judgment and evaluation on pathology teaching was made through teachers' feedback and students' questionnaire survey.Results showed that during the process of situational teaching centered on students,teaching forms were diversified,teacher evaluation was high and students' responses were active.Application of situational teaching competition made exchanging the roles of teachers and students and teaching in joy,which on the one hand stimulated students' enthusiasm,initiative and creativity and on the other hand improved teaching level of teachers.Satisfactory teaching effects were achieved after using this method.

Due to the complicated pathogenesis of cardiac arrhythmia, the safe and effective therapeutic strategies for cardiac arrhythmia remain an urgent medical problems in the recent years. In this paper, we introduced the research practice of anti-arrhythmic agents targeting on potassium ion channel. The research progress of anti-arrhythmic agents in up-to-date literatures were also reviewed and prospected.

ABSTRACT

<p><b>OBJECTIVE</b>To discuss the surgical treatment of the giant aneurysms of middle cerebral artery.</p><p><b>METHODS</b>Clinical data, surgical methods and outcomes were analyzed in 17 giant aneurysms of middle cerebral artery treated from January 2001 to March 2008. CT scan, CTA, MRA, DSA and 3D-DSA were performed before operations so that we could comprehend the location, size, and shape of aneurysms and compensatory circulation of collateral branches to design the individualized treatment options. All patients had been surgically treated mostly by modified pterional approach, of which, direct clipping of the aneurysms was accomplished in 4 patients, aneurysms trapping or removal after trapping in 4, aneurysms excision or trapping combined with vessels reconstruction in 7, and aneurysms wrapping in 2 cases.</p><p><b>RESULTS</b>CT and MRI revealed the shape and size of aneurysms clearly, while DSA and 3D-DSA could demonstrate the aneurysm's neck and relationship with the adjacent structure. Postoperative neurological function was evaluated according to Glasgow Outcome Scale when patients were discharged. Twelve patients had excellent neurological outcomes. However 4 patients were moderately disabled and one were severely disabled. No patient was dead postoperatively.</p><p><b>CONCLUSIONS</b>It is necessary to perform elaborate imaging before operations for individualized surgical planning. The temporary occlusion of the parent artery and elimination of intra-aneurysmal thrombus are helpful to clipping the aneurysmal neck. Vessels reconstruction is a new and effective method of treating the giant aneurysms of middle cerebral artery.</p>

ABSTRACT

Objective: To investigate the antihypertensive time-effect and dose-effect features of Sancao jiangya decoction(SCD).Methods: The blood pressure of spontaneously hypertensive rats at different time points were measured after treatment with Sancao jiangya decoction of low,middle,high concentration by tailartery blood pressure measurement for conscious rats.Results: The blood pressure was decreased at 2 hours after drug taken,there were significant dose-effect relationship between the antihypertensive effect and the low,middle,high dose.At 4h after drug taken,the high,middle dose had dose-effect correlation,but the low-dose had no antihypertensive effect.Further research on the middledose shows that the blood pressure reduced at 1h after drug taken,and the stable antihypertensive effect was keeping during 1-4h,the blood pressure began to rise at 6h,and got back to the level before drug taken at 8h.Conclusion: To choose the Middle-dose(10.4g crude drug/kg body weight) and 2h after drug taken is appropriate for SCD's use.This result laid a substantial foundation for further research on effects evaluation and mechanism of antihypertensive medicine.

ABSTRACT

<p><b>OBJECTIVE</b>The expression of C/EBPalpha protein and mRNA during automatically activation process in primary cultures of HSCs were observed in order to explore its possible association with the proliferation and activation of HSCs.</p><p><b>METHODS</b>Immunocytochemistry, Western blot and RT-PCR were used to evaluated the expression of C/EBPalpha protein and mRNA; as well as the expression of alpha-SMA, Desmin, MMP2, type I procollagen (alpha1). The eukaryotic vector harboring the full length cDNA of C/EBPalpha was transfected into activated HSC, then immunocytochemistry was applied to confirm the transfection and evaluate the effect of transfection on the proliferation of HSC by calculating the PCNA-positive cells. The morphological changes of HSC were observed by use of phase-contrast microscope.</p><p><b>RESULTS</b>Constitutive expression of mRNA and protein of C/EBPalpha were detected in primarily cultured HSCs, and the protein was seen in both nuclei and cytoplasm with the latter being dominant. Their expression levels reached highest at day 2 of the culture, then decreased gradually when continually cultured to the day 4, 7, 10, on the other hand, the expression of alpha-SMA, MMP2 and ColI(alpha1) increased steadily. Transient transfection was verified by the fact that much more and stronger C/EBPalpha stain was observed in transfected HSCs than in void-vector transfected cells. In C/EBPalpha gene transfected HSCs, the number of PCNA-positive cells dramatically decreased compared with the void-vector transfected cells 24h after transfection. In addition, the C/EBPalpha gene transfected HSCs died 36 h after transfection, a few surviving cells became longer and thinner in morphology, however the void-vector transfected cells almost all remained alive.</p><p><b>CONCLUSIONS</b>C/EBPalpha was likely involved in the HSCs activation, and over-expressed C/EBPalpha by transfection had inhibitory influence on the proliferation of cultured rat HSCs.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the feasibility of using iron oxide nanoparticles as gene vector and the effect of magnetic field on efficiency of transfection.</p><p><b>METHODS</b>Iron oxide nanoparticles were prepared by alkaline precipitation of divalent and trivalent iron chloride. The surface of iron oxide nanoparticles was modified by self-assembled poly-L-lysine to form particle complexes (IONP-PLL). Transfection was determined by delivering reporter gene, PGL2-control encoding luciferase, to different cell lines using IONP-PLL as vector. The effect of magnetic field on efficiency of transfection was determined using Nd-Fe-B permanent magnet.</p><p><b>RESULTS</b>Foreign gene could be delivered to various cell lines by IONP-PLL and expressed with high efficiency, but the transfection efficiency and time course varied in the different cell lines studied. Magnetic field could enhance the efficiency of transfection by 5 - 10 fold.</p><p><b>CONCLUSION</b>IONP-PLL can be used as a novel non-viral gene vector in vitro, which offers a basis for gene delivery in vivo.</p>

ABSTRACT

Objective To investigate the effects of host factors and hepatitis B virus (HBV) factors on interferon treatment in patients with chronic hepatitis B(CHB). Methods Polymerase chain reaction sequence specific primers (PCR SSP) technology was used to determine human leukocyte antigen (HLA) DR and DQ alleles in 32 patients with CHB. Serum HBV DNA level was quantified by a PCR assay with a low limit of detection of 1000 copies/ml. HBV genotypes, precore(A 1896 ) and core promoter(T 1762 A 1764 ) mutations was determined by PCR restriction fragment length polymorphism(PCR RFLP) analysis. Results The frequency of HLA DRB1*14 allele was higher in 10 responders to IFN treatment than in 22 non responders(20.0% v.s. 2.3%, relative risk(RR)=10.750, P =0.030); whereas that of HLA DQB1*07 is lower(10.0% v.s. 38.6%, RR=0.176, P =0.022). Pretreatment serum HBV DNA level in 10 responders was lower than that in 22 non responders (6.71?1.06 v.s. 7.59?0.56log 10 copies/ml, P =0.030). Patients with HBV genotype B infection had a higher response rate to IFN than those with HBV genotype C infection(53.8% v.s. 15.8%, P =0.049). Pretreatment serum ALT level, presences of A 1896 mutation and T 1762 A 1764 mutations had no difference between responders and non responders. Logistic multivariate analysis identified low pretreatment HBV DNA level(RR=0.212, P =0.034), presence of A 1896 mutation(RR=0.063, P =0.049) and HLA DQB1*07(RR=13.358, P =0.045) as independent factors associated with IFN antiviral response. Conclusions Low load of pretreatment serum HBV DNA and T 1762 A 1764 mutation in HBV genome or HBV of genotype B are associated with the response rate to IFN treatment in patients with CHB.

ABSTRACT

The effects of several factors on the chlamydospores production of Trichoderma aureoviride T-33 during the fermentation were researched.Based on the results above, the orthogonal test was made to screen out the best prescription.The results showed that, the best single-factor conditions for the chlamydospores production of T.aureoviride T-33 were, liquid culture of oat powder, 30℃, pH4.0, 120r/min, 24 hours oscillate incubating as well as liquid culture volume of 80mL/bottle when the 250mL size triangle bottle was used.The result of orthogonal test showed that, the best prescription for temperature, pH and oscillating speed was 30℃, pH4.0 and 140r/min.3.37?10~ 7 spore/mL chlamydospores were obtained at this combined condition.
